下線を引く 文字や語句の下に線を引いて、目立たせること。手書きでも、パソコンの文字装飾でも使えます。
強調する、明確に示す 事実・発言・出来事などが、ある点の重要性をはっきり示すという意味で使います。
下線、アンダーライン 文字の下に引かれた線のこと。印刷物や文書編集で使われます。

underline は文字の下に線を引くことです。highlight は蛍光ペンで目立たせることや、重要点を取り上げることを指します。比喩的な意味ではどちらも「強調する」と訳せることがあります。

文書で文字の下に線を引く場合は underline が普通です。underscore は主に「_」という記号や、比喩的に「強調する」という意味で使われます。
